pH and charge reversal-driven nanoplatform for efficient delivery of therapeutics

Nanomedicine which delivers therapeutics to tumours holds great potential for cancer treatment. However, endosomal trapping and uncontrollable release usually limit the efficiency of nanomedicine. Herein, a smart mesoporous silica based nanoplatform was constructed, in nanoparticles (MSNs) serve as core, capped with pH-induced charge-reversal polymer -PAH-cit- cationic polyelectrolyte polyethyleneimine (PEI). The oppositely charged can not only act gatekeeper controlled release, but also mediated efficient escape therapeutics. Under acidic environment, hydrolysis acid-cleavable bonds PAH-Cit would trigger charge reversal drug release. Furthermore, prepared demonstrated higher tumor cell proliferation inhibition rate than free theruputics vitro assays significantly inhibited tumour growth 4T1 model mice. Therefore, our strategy offers simple general delivery
